Profiling metabolites, lipids, and chemical exposures relies upon mass spectrometry. Although targeted applications such as newborn screening demonstrate that mass spectrometry can be scaled to millions of people, measurements of the comprehensive metabolome, lipidome, and exposome have not yet reached the same population level. Recent evidence from whole genome sequencing of subjects contributing to biorepositories such as the UK Biobank has taught us that large-scale omics profiling has the potential to yield critical new insights into health and disease. The goal of this presentation is to review the barriers hindering analogous studies in metabolomics, lipidomics, and exposomics. A workflow that overcomes these limitations will be discussed and its application to cohorts of tens of thousands of participants described. Lastly, the benefit of combining metabolomics, lipidomics, and exposomics with other omics modalities such as genomics and proteomics will be examined. A perspective will be offered for how multi-omics profiling at the population level can best advance the vision of precision medicine.
